Title: Analysis of Digitalized Microscope Images: an Approach to Study ANA Fluorescence Patterns

Abstract: The present study was aimed to perform an analysis of digitized fluorescent microscope images so that to obtain objective quantitative characteristics of different ANA fluorescent patterns. Prediluted sera from patients with organ non-specific autoimmune disorders were tested on acetone-fixed rat liver sections in indirect immunofluorescent assay (IFA). The images from homogenous, rim, and speckled fluorescence were collected using a fluorescent microscope with CCD camera attachment. The computer analysis included transformation from colour (RGB) to achromatic (HSL) image as well as various filtering procedures. In order to obtain objective parameters of the images, two types of information were developed: surface plot measurements and line density plot profiles. Statistical mean image pixel values were recorded. On the basis of these, statistical data definite quantitative evaluation was obtained for the presence of immune reaction and its localization.
